4-picolinic acid (1g, 7.74mmol) is dissolved in NaOH solution (2 M,10 mL), THF (10 mL) solution dissolved with BOC anhydride (2.53g, 11.61mmol) is slowly added under ice bath condition, the reaction is stirred at room temperature for 1 hour, THF is removed by rotary evaporator, the pH value of aqueous phase is adjusted to 5-6 with dilute hydrochloric acid, and then extracted with ethyl acetate, the combined organic phase was dried and filtered with anhydrous Na2SO4, and the filtrate was concentrated with a rotary evaporator. After vacuum drying, the white solid compound 1-Boc-4-piperidinic acid (1.78g, 7.76mmol, yield 100%) was obtained. |
